چکیده

The Hanahan protocol is widely used to transform bacterial cells with a plasmid of interest. However, little is known about the mechanism by which bacterial cells, particularly Escherichia coli, take up these plasmids. It was hypothesized that the PitA phosphate uptake system, which has been shown to transport divalent metal cations including Ca, may be involved in the Ca-mediated plasmid uptake in the Hanahan protocol. The model on which this hypothesis is based involves the binding of plasmid DNA by Ca, followed by the localization of the Ca-plasmid complex to the PitA system and subsequent complex uptake into the cell. In this study, the transformation efficiencies of a pitA mutant and a pitA wild type control were compared. A modified version of the Hanahan protocol was used to transform these strains with plasmid pBR322. Transformation efficiencies were determined by comparing ampicillin-selective with non-selective plate counts. This study indicated that the presence or absence of pitA has little impact on plasmid uptake in these transformations. Future studies on the basis of this study’s data must be done at plasmid-saturating conditions for both the mutant and parental strains in order to support these results.

Activation by gene amplification of pitB, encoding a third phosphate transporter of Escherichia coli K-12.

Two systems for the uptake of inorganic phosphate (P(i)) in Escherichia coli, PitA and Pst, have been described. A revertant of a pitA pstS double mutant that could grow on P(i) was isolated. We demonstrate that the expression of a new P(i) transporter, PitB, is activated in this strain by a gene amplification event.

Impact of heat shock step on bacterial transformation efficiency

CaCl2 treatment followed by heat shock is the most common method for artificial transformation. Here, the cells were transformed using CaCl2 treatment either with heat shock (standard protocol) or without heat shock (lab protocol) to comprehend the difference in transformation efficiency. The BL21 strain of Escherichia coli (E. coli) was being susceptible using CaCl2 treatment. Some Cells were ...

Tellurite enters Escherichia coli mainly through the PitA phosphate transporter

Several transporters suspected to be involved in tellurite uptake in Escherichia coli were analyzed. Results showed that the PitA phosphate transporter was related to tellurite uptake. Escherichia coli ΔpitA was approximately four-fold more tolerant to tellurite, and cell viability remained almost unchanged during prolonged exposure to the toxicant as compared with wild type or ΔpitB cells. Not...

Characterization of PitA and PitB from Escherichia coli.

Escherichia coli contains two major systems for transporting inorganic phosphate (P(i)). The low-affinity P(i) transporter (pitA) is expressed constitutively and is dependent on the proton motive force, while the high-affinity Pst system (pstSCAB) is induced at low external P(i) concentrations by the pho regulon and is an ABC transporter. We isolated a third putative P(i) transport gene, pitB, ...
